Optimization of Fermentation Parameters for Phytase Production by Bacillus sp. (in: firmicutes) strain MJ Isolated from Poultry Feces

Abstract
The presence of phytate in animals’ feeds restricts the availability of phosphorus and other nutrients for monogastric animals. This research is aimed to optimize the fermentation parameters for enhancement of phytase production by Bacillus sp. (in: firmicutes) strain MJ. Bacteria isolates from poultry farm house were screened for phytase production on phytase screening medium (PSM). The optimization of fermentation parameters was assessed by central composite design (CCD), while the interaction of carbon, nitrogen, pH and inoculums size were studied by response surface methodology (RSM). One isolate identified as Bacillus sp. (in: firmicutes) strain MJ (OR852745) produced the highest clear zone (7 mm). The model predicted optimum phytase production at 6.97 U/mL using 2.72 g of carbon, 1.5 g of nitrogen 0.3 mL of inoculums at pH 6.9 closed to the experimental optimal phytase of 7.27 U/mL. This condition yielded 1.29-fold increase over the unoptimized medium. The present study suggested that the phytase from Bacillus sp. (in: firmicutes) strain MJ may have a wide industrial application.
